CJC-1295 and Ipamorelin Research Guide: Growth Hormone Secretagogue Science

CJC-1295 and Ipamorelin are two of the most widely studied growth hormone secretagogues in peptide research. They work through different but complementary mechanisms, and combining them has become one of the most common research protocols in the growth hormone field.

CJC-1295: A GHRH Analog

CJC-1295 is a synthetic analog of Growth Hormone-Releasing Hormone (GHRH). GHRH is the natural signal that tells the pituitary gland to produce and release growth hormone.

How it works. CJC-1295 binds to GHRH receptors on the pituitary, stimulating growth hormone synthesis and release. The synthetic modification extends its biological half-life significantly compared to natural GHRH, which degrades in minutes.

DAC vs. no-DAC variants. CJC-1295 exists in two forms. The DAC (Drug Affinity Complex) version binds to albumin in the blood, extending its half-life to approximately 8 days. The no-DAC version (also called Modified GRF 1-29) has a shorter half-life but produces more distinct pulsatile GH release. Most research protocols use the no-DAC variant for its more physiological release pattern.

Ipamorelin: A GHRP

Ipamorelin is a Growth Hormone-Releasing Peptide (GHRP). Unlike CJC-1295, which mimics the body’s own GHRH, Ipamorelin works through the ghrelin/GHS receptor pathway.

Selectivity. What distinguishes Ipamorelin from other GHRPs (like GHRP-6 or GHRP-2) is its selectivity. Research has shown that Ipamorelin stimulates growth hormone release without significantly affecting cortisol, prolactin, or appetite hormones. This selectivity makes it particularly useful in research settings where isolating GH effects from confounding hormonal changes is important.

Concentration-dependent response. Published studies show a clear concentration-response relationship with Ipamorelin, and importantly, the GH response does not diminish with repeated exposure in the way some other secretagogues do.

Why They’re Combined

The combination of CJC-1295 and Ipamorelin is one of the best-studied peptide pairings in growth hormone research. The rationale is mechanistic:

CJC-1295 stimulates the pituitary through the GHRH pathway (the “release” signal). Ipamorelin stimulates through the ghrelin pathway (the “amplify” signal). When both pathways are activated simultaneously, the result is a synergistic increase in growth hormone release that exceeds what either compound produces alone.

Think of it as pressing the gas pedal (CJC-1295) while also removing the brake (Ipamorelin reduces somatostatin’s inhibitory effect). The combined output is greater than the sum of the parts.
